It all depends on your playing style, combined with what you are doing atm.

Trader Specialisation is very good for if you have many golden cities. But... This will come at a research disadvantage. So this would be good if you have all the research completed which you would need, and you have loads of golden city's on desert land.

Cleric Specialisation is required if you want to build up an army, and keep it alive. Every army needs healers, and lots of em. And we all know the healers are produced very fast. *cough* So use this if you need healers for ruin hunting or PvP.

Miner Specialisation is not that good for a knight, since a golden city will produce much more resources, if (!) you can buy your resources at a low price. That's why many knights build traders on the normal mines in villages.

Benefactor Specialisation can be usefull if you are building lots of soldiers for your army, or if you combine an army city with an golden city. The extra people per cottage will generate gold (taxes), and the growth rate will make sure your cities won't die out due to recruiting.

Architect Specialisation is very usefull if you've got an basic army and don't need to get healers fast. You will get lots of resources from ruin hunting, and need to spend them just as quickly. This specialisation has less value if you have big city defences (you never need to rebuild anything), and you have every building lvl 7. Or if you aren't that active...

Defender Specialisation is for turtles. What are turtles? Turtles are players who hide behind their defences. This is an must-have for the players who don't want to spend alot of time. Feeling save as to be offline a few days. (not really ideal for suzarians who go off for a few days, but if you put your army in the sanctuary... Who cares.
